A revolutionary COVID-19 (S glycoprotein) rapid test strip

A: Not for current challenges

We have developed a revolutionary COVID-19 (S glycoprotein) rapid test strip, which is completely different from nucleic acid detection and antibodies detection. 

The advantages of this new antigen (S protein) detection strip are:

1. Rapid direct detection: can detect the presence of virus directly and get the result within 5-15 minutes.

2. Non-invasive detection: innovative use of respiratory secretions (nasal mucus, sputum) and stool testing,.

3. High specificity: the use of a new generation of detection method, more specific than the traditional strip, better sensitivity, no cross-reaction..

4. Fearless virus mutation: can directly detect SARS- cov-2 and all mutated virus strains.

5. Multi-purpose: it can be used for home detection, detection of designated institutions such as hospitals, screening before returning to work in enterprises or school; It can be used directly for qualitative testing, or for quantitative analysis with a handheld auxiliary testing equipment.

Have you validated this method, if so, how and what were the results of the validation?​

We have obtained international patents(PCT) and CE certifications, and as the world's only company developed for COVID - 19 antigen rapid test strip, our products have been included in the FIND (WHO Collaborating Centre for Laboratory Strengthening and Diagnostic Technology Evaluation) list.

 

How quickly could this be deployed and what are the dependencies?​

The role of ACE2 in SARS-CoV-2 infection. Studies have shown that S protein of SARS-CoV-2 has a strong binding affinity with human ACE2 receptor, KD=15nM, which is 10-20 times stronger than SARS virus. This may be one reason why SARS-CoV-2 is so contagious.

We established an chromatography assay to demonstrate that the S protein of novel coronavirus has a stronger binding force with human ACE2 receptor in human respiratory secretions than in PBS (pH7.4) .

On the strip's nitrocellulose (NC) membrane, the T line is coated with the recombinant SARS-CoV-2 S1 protein subunit, and the control (C) line is coated with goat anti-rabbit IgG polyclonal antibodies. Quantum dot–labeled ACE2 protein and rabbit IgG are embedded in the reagent pad.

During testing, when three drops of the sample are added, the sample laterally flows from bottom to top due to capillary action. If the sample does not contain the virus, the quantum dot–labeled ACE2 protein is captured and bound by the S1 protein coated onto the T line. If the sample contains the virus, the quantum dot–labeled ACE2 protein is neutralized by the virus’s S protein, after which the level of ACE2 captured by the recombinant S1 protein coated onto the T line will decrease.

150,000-200,000 test / day

What are the risks and barriers to using this at scale?​

No other risks and obstacles were found. On the contrary, many originally limited problems have been better resolved.

How does this fit into a tracked sample pathway?

Qualitative detection can be done using ultraviolet flashlight. It can also be inserted into handheld auxiliary testing equipment for quantitative analysis.

All quantitative analysis data can be exported, summarized and analyzed for epidemiological analysis and research.
